From my experience, Tiagra shifters do not work well with the Shimano Ultrega RD-6600 10-speed rear derailleur, on the contrary, they work perfectly with the front derailleur.

It is not compatible with the rear derailleur. It is compatible with the front derailleur.

It depends on what size cassette you are running in the back and what size chainrings you have got up from. As I have gotten a bit older, I needed a bigger cassette on the back of my 2006 Cannondale Synapse carbon. The bike had the full Dura Ace 7800 groupset. I went to a Tiagra 12-34 cassette and a Tiagra rear derailleur but then the old Dura Ace shifters did not index properly. My bike shop told me I had to get the Tiagra Brake Lever Set with Shifter (listed above). The old Dura Ace front derailleur works fine but I had to get the Tiagra rear derailleur in order to get it to work with the larger cassette. So, I would suggest asking your bike shop, but my suspicion is that your rear derailleur might work pretty well as long as you are not running more than an 11-28 cassette. Even then, the indexing might not be perfect. Based on my experience I would keep your existing front derailleur but most likely replace your Ultegra rear derailleur with a Tiagra rear derailleur. I am very happy with the performance of the Tiagra components. Thank you trickle down technology.

It is compatible with from derailleur, but the cable pull ratio is slightly different with ultegra rear derailleur, would be better shifting if you used a 4700 10 speed Shimano rear derailleur

It comes with both and all the cables and housing. Just a heads up that it might not work well with your older Ultegra. I would get the frond and rear derailleurs to make sure it would work well.
